TTI1 also known as TEL2-interacting protein 1 is a protein weighing approximately 245 kDa. It is expressed widely across various tissues including liver kidney and brain. Mechanically TTI1 operates as an important scaffolding protein that supports the formation of multiprotein complexes specifically those involved in processes of cell growth and proliferation. TTI1 interacts with other proteins to execute these structural roles facilitating precise coordination required for cellular machinery and function.
Biological function summary
TTI1 is a part of the larger complex known as the TTT complex which consists of TTI1 TELO2 and TTI2 proteins. This protein complex stabilizes another set of critical proteins called PIKKs which include mTOR ATM and ATR. These proteins play roles in regulating cell growth DNA damage response and metabolism. By participating in this complex TTI1 plays a role in maintaining cell integrity by interfacing with cellular stress responses and ensuring proper progression and repair of the cell cycle.
Pathways
TTI1 integrates functions within the mTOR signaling pathway and the DNA damage response pathway. In the mTOR pathway TTI1 influences nutrient signaling and energy balance involving cooperation with mTOR which modulates cell metabolism and proliferation. In the DNA damage response pathway the interactions with ATM and ATR are pivotal as they control aspects of DNA repair processes and cellular checkpoints ensuring genomic stability. These pathways illustrate how TTI1 manages to link nutrient signaling with genome maintenance.
TTI1 has associations with cancer and neurodegenerative diseases. In cancer the interactions with mTOR and ATM pathways often result in aberrant signaling leading to uncontrolled cell proliferation. Evidence suggests that these signaling disruptions may contribute to tumorigenesis and treatment resistance. In neurodegenerative diseases disruptions in TTI1-mediated pathways may result in impaired stress responses and cellular repair mechanisms potentially accelerating neuronal cell damage and death. Understanding these relationships highlights the importance of TTI1 in both cellular physiology and pathology.
