Product details:
Human VEGFC ELISA Kit ab100664 is a sandwich ELISA to measure Human VEGFC in serum, plasma, cell culture supernatant with a sensitivity of 15 pg/ml.
How the assay works
This assay employs an antibody specific for Human VEGFC coated on a 96-well plate. Standards and samples are pipetted into the wells and VEGFC present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human VEGFC antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of VEGFC bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

1. Add standard or sample to each well used. Incubate at room temperature
2. Wash and add prepared biotinylated antibody to each well. Incubate at room temperature.
3. Wash and add prepared streptavidin solution. Incubate at room temperature.
4. Add TMB Development Solution to each well. Incubate at room temperature.
5. Add Stop Solution to each well. Read at 450nm immediately.
Optimization may be required with urine samples.

VEGFC also known as Vascular Endothelial Growth Factor C is a member of the VEGF family involved in angiogenesis and lymphangiogenesis. It has a molecular mass of approximately 35 kDa. VEGFC is expressed in various tissues with high levels in the heart lung and muscle. It acts primarily through its interaction with the VEGFR-3 receptor stimulating the formation of blood and lymphatic vessels. This protein plays an important role in the development and maintenance of the circulatory and lymphatic systems.
Biological function summary
VEGFC contributes to the regulation of vascular and lymphatic endothelial cell functions. It is not known to be part of a larger protein complex but exerts its functions through interactions with receptors such as VEGFR-3. By promoting cell proliferation and migration VEGFC assists in the repair and regeneration of tissues. Its role in fluid balance is essential as it helps maintain tissue equilibrium and immune function.
Pathways
VEGFC is integral to the VEGF signaling and lymphatic vessel development pathways. Within these processes VEGFC works alongside other proteins such as VEGFA and VEGFB to modulate vascular permeability and vessel formation. The interaction between VEGFC and the VEGFR-3 receptor specifically mediates lymphatic endothelial cell activation and function linking it to vital physiological processes like inflammation and tissue repair.
VEGFC is associated with cancer and lymphedema. Its role in promoting angiogenesis is critical in tumor growth and metastasis with its overexpression often linked to cancer progression. Furthermore insufficient VEGFC activity contributes to the development of lymphedema due to inadequate lymphatic drainage. In cancer pathology VEGFC works together with VEGFR-3 contributing to the abnormal growth of lymphatic vessels which in turn facilitates cancer cell dissemination.
