SIGLEC11 also known as sialic acid-binding Ig-like lectin 11 is an inhibitory receptor protein characterized by its ability to bind sialic acids. It has a mass of approximately 66 kDa. This protein contains an immunoreceptor tyrosine-based inhibitory motif (ITIM) that helps transmit inhibitory signals. SIGLEC11 is predominantly expressed in the immune cells including macrophages and dendritic cells within the central nervous system (CNS). It is also found in certain peripheral tissues.
Biological function summary
SIGLEC11 plays a role in modulating immune responses and maintaining immune homeostasis. By binding to sialic acid-containing glycoproteins SIGLEC11 can influence cell-cell interactions and prevent unnecessary activation of immune cells. It does not form part of a larger complex but directly participates by recruiting phosphatases such as SHP-1 or SHP-2. These recruitments help dephosphorylate substrates in turn dampening immune cell activation.
Pathways
SIGLEC11 contributes to the negative regulation of receptor signaling pathways relevant to the immune system. Within the signaling pathways it impacts the downstream signaling cascade that involves Src family kinases. SIGLEC11 shares interactions with other members of the SIGLEC family engaging in pathways that modulate cellular activation and inflammation such as the MAPK pathway. This ensures balanced immune responses to avoid excessive inflammation.
SIGLEC11 expressions show relevance to neurodegenerative diseases and autoimmune disorders. Altered SIGLEC11 activity is associated with Alzheimer's disease where it may dampen the inflammatory response within the CNS providing a protective mechanism. In autoimmune conditions such as systemic lupus erythematosus (SLE) SIGLEC11 may interact with proteins that regulate self-tolerance and immune activation mitigating inappropriate immune responses.
