ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate qualitative measurement of IgM class antibodies against Brucella in Human serum and plasma.
A 96-well plate has been precoated with Brucella antigens to bind cognate antibodies. Controls or test samples are added to the wells and incubated. Following washing, a horseradish peroxidase (HRP) labelled anti-Human IgM conjugate is added to the wells, which binds to the immobilized Brucella-specific antibodies. TMB is then catalyzed by the HRP to produce a blue color product that changes to yellow after adding an acidic stop solution. The density of yellow coloration is directly proportional to the amount of Brucella IgM sample captured in plate.

Brucella IgM is an immunoglobulin associated with the early immune response to Brucella infection. Also known as Immunoglobulin M IgM this antibody is the first responder during the acute phase of infection. IgM antibodies are larger in size compared to other antibodies having a mass of approximately 900 kDa due to their pentameric structure. They are typically expressed on the surface of B cells and are found in the bloodstream. The presence of Brucella IgM indicates initial immune activation against Brucella bacteria which can cause brucellosis in humans.

Brucella IgM serves as an essential component of the body's defense mechanism against Brucella species. It plays a major role in the immune response by recognizing and binding to the Brucella antigen effectively marking these pathogens for destruction by other immune cells. As part of the immune complex IgM functions are integral in neutralizing and agglutinating pathogens.
