ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate qualitative measurement of IgM class antibodies against Tick-borne encephalitis virus in Human serum and plasma.
A 96-well plate has been precoated with Tick-borne encephalitis antigens to bind cognate antibodies. Controls or test samples are added to the wells and incubated. Following washing, a horseradish peroxidase (HRP) labelled anti-Human IgM conjugate is added to the wells, which binds to the immobilized Tick-borne encephalitis-specific antibodies. TMB is then catalyzed by the HRP to produce a blue color product that changes to yellow after adding an acidic stop solution. The density of yellow coloration is directly proportional to the amount of Tick-borne encephalitis IgM sample captured in plate.

The target Tick-borne encephalitis (TBE/FSME) IgM plays a critical role in the immune response during the acute phase of TBE infection. TBE IgM antibodies are part of the immunoglobulin M class which the body first produces in response to an antigen. Having a pentameric structure each IgM molecule has a mass of approximately 970 kDa. These antibodies circulate in the bloodstream and appear at the onset of symptoms serving as a marker for recent infection by the TBE virus which is a member of the flavivirus family.
Biological function summary
These IgM antibodies aid in neutralizing the flavivirus particles and preventing them from infecting more host cells. Their activity helps to control the viral load within the host and their presence indicates an active immune response. IgM antibodies do not form part of a larger protein complex instead they function independently to bind specific antigens. Their ability to form multivalent antigen-antibody complexes enhances the efficiency of pathogen neutralization.
Pathways
The TBE/FSME IgM plays a role in the classical complement activation pathway. This pathway involves a series of protein interactions that lead to the destruction of pathogens. During TBE infection the activated complement cascade enhances the opsonization and clearance of the virus. The pathway also interacts with other immunoglobulins and various proteins within the complement system such as C1q which binds to the Fc region of IgM initiating the classical pathway.
The presence of TBE IgM antibodies is a significant marker for diagnosing tick-borne encephalitis. TBE is a neurological condition caused by the TBE virus often resulting in inflammation of the brain and central nervous system. The detection of IgM indicates a current or recent viral infection. Additionally these antibodies play a role in differential diagnosis helping to distinguish TBE from other flavivirus infections such as West Nile virus and their associated unique protein interactions.
