E2F8 also known as transcription factor E2F8 plays a vital role in cell cycle regulation. This protein has a molecular weight of approximately 97 kDa. E2F8 is widely expressed in various tissues with significant levels detected in the liver and placenta. As part of the E2F family it functions as a transcription factor repressing or promoting the transcription of target genes involved in cell proliferation.
Biological function summary
E2F8 acts as a regulator within the context of the cell cycle. It forms a complex with other E2F family members such as E2F7 which can bind to specific DNA sequences and regulate the transcription of genes necessary for the G1/S transition. This activity makes E2F8 essential for controlling cell growth and maintaining cellular homeostasis. E2F8's ability to act as a repressor distinguishes it from other E2Fs which typically activate transcription.
Pathways
E2F8 integrates into the mechanistic framework of the cell cycle and DNA repair pathways. Its activity is closely tied to the p53 pathway where it can influence p53-mediated transcriptional repression. E2F8 influences other family proteins like E2F1 which are important in driving cell cycle progression and apoptosis. By modulating these pathways E2F8 fine-tunes the balance between cell division and cell death—an important aspect of normal development and tumor suppression.
E2F8 has a notable connection with cancer and developmental disorders. Its dysregulation has been observed in various cancers such as hepatocellular carcinoma where it can promote oncogenic transformation by altering cell cycle dynamics. E2F8 also interacts with proteins like CCNE1 which is often overexpressed in tumors. Furthermore abnormalities in E2F8 function can contribute to birth defects by disrupting normal cell cycle control during embryonic development.
