RHOH also known as Rho-related GTP-binding protein RhoH is a small GTPase belonging to the Ras superfamily. It has a molecular weight of approximately 21 kDa. This protein is expressed mainly in hematopoietic cells such as lymphocytes where it plays a mechanical role in modulating cytoskeletal organization by regulating actin filament structures. Unlike other Rho GTPases RHOH lacks the intrinsic GTPase activity which means it stays in the GTP-bound form and functions as a constitutive signaling molecule.
Biological function summary
RHOH contributes to signal transduction pathways essential for immune function. It is involved in T-cell receptor signaling and acts as a negative regulator of the RhoA and Rac1 pathways. RHOH forms complexes with other signaling molecules which impacts T-cell development and activation linking cytoskeletal dynamics and membrane trafficking to signaling processes.
Pathways
Researchers identified RHOH as significant in the NF-kB signaling pathway and T-cell antigen receptor (TCR) signaling. Through these pathways RHOH interacts with proteins like ZAP-70 and Lck influencing the downstream signaling events important for immune cell activation and response. Consequently RHOH modulates interactions that are vital for immune system adaptations and activities.
RHOH mutations and dysregulation connect to specific forms of leukemias and lymphomas. Aberrant expression or function of RHOH correlates with chronic lymphocytic leukemia (CLL) wherein altered signaling contributes to cancer pathogenesis. Through these disease states RHOH is linked with proteins such as Syk which also play roles in the altered signaling pathways characteristic of these cancers.
