Product details:
The Human Stanniocalcin 1 / STC Enzyme-Linked Immunosorbent Assay (ELISA) kit (STC1) (ab213829) is designed for the quantitative measurement of Human Stanniocalcin 1 / STC in cell culture supernatants, cell lysates, serum and plasma (heparin, EDTA).
The ELISA kit is based on standard sandwich enzyme-linked immunosorbent assay technology. A monoclonal antibody from mouse specific for Stanniocalcin 1 / STC has been pre-coated onto 96-well plates. Standards (NSO, V34-A247) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Stanniocalcin 1 / STC is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB is used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human Stanniocalcin 1 / STC amount of sample captured in plate.
Stanniocalcin 1 / STC is a glycoprotein which is encoded by the STC1 gene. This gene encodes a secreted homodimeric glycoprotein that is expressed in a wide variety of tissues and may have autocrine or paracrine functions. Human Stanniocalcin 1 / STC is a putative molecular biomarker of leukemic microenvironment and the only molecular function known up to date is a SUMO E3 ligase activity in the SUMOylation cycle. Stanniocalcin 1 / STC interacts with lots of proteins in the cytoplasm, mitochondria, endoplasmatic reticulum and dot-like fashion in the nucleus. The N-terminal region of Stanniocalcin 1 / STC is the function region which is responsible to establish the interaction with its partners, including SUMO1. Low resolution studies show that Stanniocalcin 1 / STC is an anti-parallel homodimer in solution and the cysteine 202 is responsible for the dimerization of this protein. All the 5 disulfide bonds of human STC1 are conserved and have the same profile of fish STC.

Stanniocalcin 1 (STC1) is a glycoprotein hormone with a mass of about 56 kDa. Alternate names for this protein include STC and STC-1. It is expressed in various tissues such as the kidney brain and ovary. STC1 plays a significant mechanical role in the regulation of calcium and phosphate transport across cell membranes. It interacts with certain receptors on cell surfaces to modulate ion exchange contributing to the maintenance of mineral homeostasis.
Biological function summary
Stanniocalcin 1 regulates calcium and phosphate balance and performs roles in cellular metabolism and antioxidant defense. It acts independently not as part of a complex but sometimes affects cascades leading to oxidative stress responses. STC1 adjusts energy metabolism by managing reactive oxygen species thereby enhancing cell survival under stress conditions. Its signaling impacts energy balance and adaptive responses in metabolically active tissues.
Pathways
Despite its independent operation Stanniocalcin 1 is integrated into oxidative stress and inflammatory pathways. It is involved in the Akt/PKB signaling pathway which influences cell survival and stress response. STC1 intersects with other proteins like PTH (parathyroid hormone) within the calcium metabolism pathways affecting bone and renal function. Through these pathways it facilitates cellular adaptation to metabolic demands.
Researchers link Stanniocalcin 1 to osteoporosis and cancer. Its role in calcium homeostasis makes it relevant in osteoporosis where bone density and strength are compromised. In the context of cancer STC1's influence on cell survival and stress responses may contribute to tumorigenesis although its function can be protective or detrimental depending on the cellular environment. The interaction of STC1 with proteins like PTH highlights its involvement in bone-related pathologies and potential roles in cancer progression.
