Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Pro341 of human respiratory syncytial virus A (strain A2) nucleoprotein (UniProt #P03418).
Background
The respiratory syncytial virus (RSV), also known as human orthopneumovirus, is a negative-sense, single-stranded RNA virus that causes serious acute respiratory disease, primarily in infants and older adults. The RSV genome consists of 10 genes encoding 11 proteins. NS1 and NS2 are nonstructural proteins. Structural proteins are matrix (M), small hydrophobic (SH), attachment glycoprotein (G), fusion glycoprotein (F), transcriptional processivity factor (M2-1), RNA regulatory factor (M2-2), nucleoprotein (N), phosphoprotein (P), and large polymerase (L). The nucleocapsid is surrounded by a host-derived lipid envelope with protruding viral glycoproteins to form the virion (1,2). N coats and stabilizes the viral RNA, and is required for replication (3). Therapeutics targeting several viral proteins are in development, including monoclonal antibodies targeting F and G proteins, and small-molecule inhibitors targeting N and L proteins (4).
