Product Usage Information Immunohistochemistry (Paraffin): 1:100 - 1:400 Storage Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at -20°C. Do not aliquot the antibody. Non-specific staining was observed in some neuronal tissue. Species Reactivity: Human, Mouse Source / Purification Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human POFUT1 protein. Background Notch signaling is essential for lymphocyte development and myelopoiesis (1,2,4,6). Notch receptors are modified by O-fucosylation catalyzed by the enzyme protein O-fucosyltransferase 1 (POFUT1) (1,2,4). POFUT1 fucosylates epidermal growth factor (EGF)-like domains found at the cell surface and in secreted glycoproteins, including Notch and its ligands. POFUT1 catalyzes the reaction by attaching fucose via O-glycosidic linkage to conserved serine or threonine residues in EGF-like domains (2,7). This particular Notch modification is critical for development and cell fate; POFUT1 deficiency leads to aberrant signaling and human disease (3,5,8,9). In addition, mouse embryonic stem cells lacking POFUT1 have normal levels of Notch receptors at the cell surface, but these receptors do not bind Notch ligands or display proper Notch signaling (4,6).
