This antibody detects exogenously expressed S-tagged proteins in cells expressed under a CMV promoter. Expression under other promoters has not been evaluated. The chimeric antibody retains its antigen-binding Fab regions from the original rabbit monoclonal antibody but contains a horse-derived Fc domain. When multiplexing, Fc-directed rabbit secondaries are required to detect rabbit-host primary antibodies. The parent antibody, S-Tag (D2K2V) XP Rabbit mAb #12774, is produced by immunizing animals with a synthetic S-Tag peptide. Background Epitope tags are useful for the labeling and detection of proteins using immunoblotting, immunoprecipitation, and immunostaining techniques. Because of their small size, they are unlikely to affect the tagged protein's biochemical properties. S-Tag is an epitope tag composed of a 15 residue peptide that, together with S-protein, comprises pancreatic ribonuclease S (1).
