Product Usage Information For optimal ChIP results, use 10 μL of antibody and 10 μg of chromatin (approximately 4 × 10 6 cells) per IP. This antibody has been validated using SimpleChIP ® Enzymatic Chromatin IP Kits. Chromatin IP: 1:50 Storage Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at -20°C. Do not aliquot the antibody. Species Reactivity: Human Source / Purification Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Tyr57 of human ZNF366 protein. Background Dendritic cells (DCs) are the watchmen of the immune system, detecting a wide variety of pathogens and initiating robust immune responses. To respond to highly variable challenges, DCs have diversified into multiple phenotypical, anatomical, and functionally distinct cell types (1,2). Zinc finger protein 366 (ZNF366) was originally identified in a yeast two-hybrid screen for transcriptional regulators responding to estrogen (3,4). ZNF366 (dendritic cell (DC)-specific transcript, DC-SCRIPT) is a member of the Cys-Cys:His-His subfamily of zinc finger proteins (5). ZNF366 localizes to the nucleus and does not show transcriptional activation via transactivation assays but does interact with co-repressors (2,5). ZNF366 has been shown to be DC-specific and is expressed in myeloid DCs, plasmacytoid DCs, and Langerhans cells (5). ZNF366 knockout mice show an immune deficiency phenotype, with substantially decreased expression of the transcription factor IRF-8 (6). Loss of function in ZNF366 has been implicated in tumor cell evasion of immune responses (7). ZNF366 represents a novel DC marker that could be involved in transcriptional repression (1,5).
