Human MMP13 ELISA Kit ab100605 is a sandwich ELISA to measure Human MMP13 in plasma, cell culture supernatant with a sensitivity of 6 pg/ml.
How the assay works
This assay employs an antibody specific for Human MMP13 coated on a 96-well plate. Standards and samples are pipetted into the wells and MMP13 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human MMP13 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of MMP13 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

1. Add standard or sample to each well used. Incubate at room temperature
2. Wash and add prepared biotinylated antibody to each well. Incubate at room temperature.
3. Wash and add prepared streptavidin solution. Incubate at room temperature.
4. Add TMB Development Solution to each well. Incubate at room temperature.
5. Add Stop Solution to each well. Read at 450nm immediately.
Optimisation may be required with urine samples.

Matrix metallopeptidase 13 (MMP-13) also called collagenase 3 is an enzyme with an approximate molecular mass of 54 kDa. It plays a significant role in the breakdown of extracellular matrix components particularly collagen. MMP-13 is expressed in several tissues including cartilage skin and bone. Its expression sees an increase during tissue remodeling and in pathological conditions.
