Product details:
Mouse Prolactin ELISA Kit ab100736 is a sandwich ELISA to measure Mouse Prolactin in serum, plasma, cell culture supernatant, urine with a sensitivity of 30 pg/ml.
How the assay works
This assay employs an antibody specific for mouse Prolactin coated on a 96-well plate. Standards and samples are pipetted into the wells and Prolactin present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse Prolactin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Prolactin bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Assay Specificity
Our ELISA kits are rigorously validated to ensure the highest level of consistency and reproducibility. Please check the protocol booklet for more details

Prolactin also known as PRL is a protein hormone with a molecular mass of approximately 23 kDa. It is primarily produced in the anterior pituitary gland but is also found in other tissues such as the placenta mammary glands and immune cells. Mechanically Prolactin functions as a peptide hormone and exerts its effects by binding to the Prolactin receptor a member of the cytokine receptor superfamily. This binding activates the Jak-STAT signaling pathway which is important for mediating the hormone's effects in target cells.
Biological function summary
The functions of this hormone extend beyond its well-known role in lactation. It helps stimulate milk production in the mammary glands by promoting the proliferation and differentiation of mammary epithelial cells. Additionally Prolactin influences reproductive functions metabolism and immune regulation. It is not part of a larger protein complex but interacts with various proteins to perform its functions. Notably it modulates the immune response contributing to tolerance during pregnancy and affecting the balance between pro-inflammatory and anti-inflammatory cytokines.
Pathways
Prolactin plays important roles in the Jak-STAT and MAPK signaling pathways. In these pathways it shares interactions with related proteins such as Growth Hormone and Prolactin Receptor aiding in cell growth differentiation and metabolism regulation. Prolactin's involvement in these pathways highlights its central role in physiological processes linking various hormonal signals to regulate body functions efficiently.
Prolactin's imbalance can lead to conditions such as hyperprolactinemia and prolactinomas both characterized by excessive levels of this hormone in the blood. These conditions are often related to alterations in the pituitary gland function. Prolactin interacts closely with Dopamine a neurotransmitter that inhibits its secretion influencing both its production and release. Dysregulation of this interaction may contribute to the development of these disorders impacting endocrine health and overall well-being.
