Product details:
Mouse SDF1 ELISA (Enzyme-Linked Immunosorbent Assay) kit (ab100741) is an
in vitro
enzyme-linked immunosorbent assay for the quantitative measurement of mouse SDF1 in serum, plasma and cell culture supernatants. (Mouse SDF1 concentration is pretty low in normal serum/plasma, it may not be detected in this assay).
This assay employs an antibody specific for mouse SDF1 coated on a 96-well plate. Standards and samples are pipetted into the wells and SDF1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse SDF1antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of SDF1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

SDF1 alpha also known as CXCL12 is a chemokine with a molecular mass of approximately 8 kDa. It plays a significant role in the immune system by directing the movement of cells through chemotaxis. This protein is predominantly expressed in various tissues including bone marrow lymph nodes and lungs. Its expression occurs across different cell types such as endothelial cells fibroblasts and stromal cells making it an essential component of the cell signaling environment.
Biological function summary
SDF1 alpha participates in regulating the movement and positioning of hematopoietic stem cells supporting tissue homeostasis and repair. It operates often by binding to the G-protein coupled receptor CXCR4. This interaction is especially important in hematopoiesis vasculogenesis and neuron guidance. Although SDF1 alpha does not function as part of a larger complex its interaction with CXCR4 plays an important role for processes like development and wound healing.
Pathways
SDF1 alpha is heavily involved in the chemokine signaling pathway which is pivotal for immune responses and cellular communication. Its interaction with CXCR4 also links it to the MAPK/ERK pathway contributing to cell proliferation differentiation and survival. These pathways highlight the essential interplay between SDF1 alpha and proteins such as CXCR4 and CCR7 which are part of broader cellular responses.
SDF1 alpha has notable implications in cancer and HIV. Its interaction with CXCR4 is important in tumor growth and metastasis influencing the tumor microenvironment. For HIV SDF1 alpha and its receptor CXCR4 are critical in viral entry into host cells. A deeper understanding of these relationships can aid in identifying therapeutic targets and diagnosing these conditions effectively.
