SLC5A3 also referred to as SMIT1 is a sodium-dependent myo-inositol transporter. It facilitates the transport of myo-inositol across cell membranes. The protein has a molecular mass of approximately 78 kDa. SLC5A3 is mainly expressed in numerous tissues including the brain kidneys and placental tissue. The transport activity is essential because myo-inositol is critical for cellular osmotic balance and signaling pathways.
Biological function summary
SLC5A3 plays an important role in osmoregulation contributing to the adaptation of cells to osmotic stress. This transporter is not part of a large protein complex yet it collaborates closely with other cellular components to maintain the inositol concentration important for phosphatidylinositol synthesis. Phosphatidylinositol serves as a precursor for inositol triphosphate which participates in intracellular signal transduction processes.
Pathways
SLC5A3 impacts the phosphatidylinositol signaling pathway and osmoregulatory mechanisms. The phosphatidylinositol signaling pathway involves other proteins like PI3K which is vital for transmitting signals that regulate various cellular processes such as growth and survival. In osmoregulation SLC5A3 adjusts the myo-inositol levels to maintain cellular homeostasis during osmotic stress conditions ensuring cell viability and function.
SLC5A3 has been linked to disorders like osmotic demyelination syndrome and bipolar disorder. In osmotic demyelination syndrome a disturbance in osmotic balance due to improper function of SLC5A3 may lead to myelin sheath damage. Additionally changes in phosphatidylinositol signaling involving SLC5A3 and interacting proteins such as GSK3β have been implicated in bipolar disorder suggesting SLC5A3 could be a potential target for therapeutic interventions in these conditions.
