Abcam's CTLA4 Mouse ELISA (Enzyme-Linked Immunosorbent Assay) kit is an
in vitro
enzyme-linked immunosorbent assay for the quantitative measurement of mouse CTLA4 in serum, plasma and cell culture supernatants.
This assay employs an antibody specific for Mouse CTLA4 coated on a 96- well plate. Standards and samples are pipetted into the wells and CTLA4 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse CTLA4 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CTLA4 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

CTLA-4 also known as Cytotoxic T-Lymphocyte Antigen 4 or CD152 is a protein receptor with an approximate molecular weight of 25–37 kDa. Expressed primarily on the surface of T cells CTLA-4 acts as an immune checkpoint molecule. It competes with the costimulatory receptor CD28 for binding to CD80/CD86 ligands on antigen-presenting cells. This competitive binding results in an inhibitory signal that reduces T cell activation and proliferation.
Biological function summary
CTLA-4 modulates immune responses by serving as a critical negative regulator in T cell activation. Unlike CD28 which promotes T cell responses CTLA-4 inhibits these responses helping to maintain immune homeostasis. CTLA-4 exists in a monomeric form on the cell surface but can form a dimeric complex when interacting with ligands. By dampening the immune response CTLA-4 prevents excessive tissue damage during immune reactions.

It functions within the context of the immune synapse influencing the balance between activation and inhibition of T cells.
