SIPA1L3 also known as Signal-Induced Proliferation-Associated 1 Like 3 or SPAR3 is a protein with a molecular mass of approximately 234 kDa. Mechanically SIPA1L3 functions as a Rap GTPase-activating protein (GAP) that regulates the activity of small G proteins by promoting the conversion of their active GTP-bound state to an inactive GDP-bound state. SIPA1L3 is widely expressed in several tissues with notable expression in the brain lens and retina indicating its involvement in neuronal development and visual processes.
Biological function summary
This protein is vital for signal transduction involved in cellular adhesion and cytoskeleton organization interacting with actin filaments to regulate cell shape and movement. SIPA1L3 is part of a multiprotein complex that influences intercellular junction formation contributing to the maintenance of cellular contacts in epithelial tissues. Its regulatory role in cellular dynamics suggests an impact on processes such as migration and proliferation.
Pathways
SIPA1L3 engages in the regulation of the Ras signaling pathway and the Rap1 signaling pathway which are important for cell growth survival and differentiation. Through its activity SIPA1L3 modulates interactions with related proteins such as Ras further influencing downstream signaling cascades. The interplay with these pathways positions SIPA1L3 as an important regulator of cellular responses to extracellular signals.
Altered expression or function of SIPA1L3 has been linked to conditions such as cataracts and neurological disorders. The misregulation of this protein can disrupt normal cellular adhesion and cytoskeleton dynamics leading to disease development. Additionally studies suggest that interactions with proteins in the Ras pathway may further contribute to its role in these diseases highlighting the relevance of SIPA1L3 in pathological states.
