CENPI also known as centromere protein I is part of the CENP family and plays a mechanical role in centromere assembly and function. It has a molecular mass of approximately 97 kDa. CENPI is expressed in various tissues but it shows higher expression levels in proliferating cells especially during cell division. It localizes to the centromere region of chromosomes and is essential for the proper functioning of the kinetochore the protein structure on chromosomes where spindle fibers attach during cell division.
Biological function summary
The role of CENPI involves its participation in the CENP-A nucleosome-associated complex. This complex is essential for the recruitment of other kinetochore proteins ensuring accurate chromosome segregation during mitosis. The proper functioning of CENPI contributes to maintaining genomic stability as errors in chromosome segregation can lead to aneuploidy which is an abnormal number of chromosomes in cells. Therefore CENPI acts as an important mediator in the accurate distribution of chromosomes during cell division.

It aids in the activation and stability of proteins like CENP-H and CENP-K which cooperate during the early stages of centromere and kinetochore formation.
