STON2 also known as stonin 2 is a protein with a molecular weight of approximately 135 kDa. It plays a role in the regulation of endocytosis by interacting with adaptor proteins involved in the internalization of synaptic vesicles. STON2 is mainly expressed in the brain where it contributes to the recycling of synaptic vesicle membranes. Its action ensures effective neurotransmitter release which is necessary for neuronal communication.
Biological function summary
STON2 functions as an important component in the synaptic vesicle cycle particularly in the clathrin-mediated endocytosis pathway. It associates with other proteins like adaptor protein 2 (AP-2) to form a complex essential for vesicle recycling. This interaction is key for the rapid turnover of synaptic vesicles enabling the nervous system's rapid response to stimuli.
Pathways
The role of STON2 in clathrin-mediated endocytosis connects it to the broader neurotransmitter release pathway. It operates alongside proteins such as synaptotagmin and synaptobrevin integral in calcium-triggered exocytosis. STON2 facilitates proper vesicle retrieval and restoration therefore maintaining synaptic integrity and efficient signal transmission.
STON2 has been implicated in neurological conditions such as epilepsy and autism spectrum disorders. Dysfunction in STON2's role in vesicle cycling can disrupt synaptic transmission potentially contributing to these disorders. Interactions with proteins like dynamin a GTPase responsible for vesicle fission further link STON2 to pathologies associated with synaptic dysfunction.
