GPCR GPR78 also known as orphan G-protein coupled receptor 78 is a member of the GPCR family which is characterized by its seven transmembrane domains. The molecular mass of GPR78 is approximately 42 kDa. It is expressed mainly in the brain with significant levels found in regions like the cortex and hypothalamus. Although research on GPR78 is still emerging it shows homology with other GPCR family members playing a role in various signaling processes triggered by interacting with extracellular ligands.
Biological function summary
GPR78 functions within the nervous system where it contributes to neurodevelopmental processes and synaptic signaling. Researchers have not yet identified specific ligands classifying it as an orphan receptor. Studies indicate GPR78's involvement in forming signaling complexes involving other membrane proteins allowing it to influence synaptic plasticity and neuron communication. This aligns with its expression in the brain hinting at its role in central nervous system activities.
Pathways
GPR78's function integrates into critical signaling cascades in the brain. It is associated with the cAMP signaling pathway which is important in neurotransmitter release and neuronal communication. The presence of GPR78 in this pathway suggests interactions with proteins like adenylyl cyclase influencing downstream signaling events. Additionally GPR78 might interact with proteins involved in the Wnt signaling pathway a fundamental process in brain development and synapse formation.
GPR78 shows potential involvement in neurological conditions specifically autism spectrum disorder (ASD) and schizophrenia. Altered expression or function of GPR78 might contribute to imbalances in neural signaling pathways with consequences for cognitive and behavioral functions. Studies have examined its interaction with proteins such as dopamine receptors which are key in these disorders. Further exploration of GPR78 might provide insights leading to novel therapeutic approaches for these conditions.
