PREX2 also known as PIP3-dependent Rac exchanger 2 acts as a guanine nucleotide exchange factor (GEF) specifically regulating the small GTPase Rac1. The PREX2 protein has a mass of approximately 212 kDa and is expressed abundantly in the brain but can also be found in other tissues. It facilitates the switch of Rac1 from its inactive GDP-bound state to the active GTP-bound state thereby modulating cell signaling processes related to actin cytoskeleton reorganization and cell migration.
Biological function summary
PREX2 significantly influences cell movement and growth. It operates as part of a multi-protein complex that coordinates various cellular signaling pathways. In cell migration and invasion PREX2 collaborates with other GEFs to promote Rac1 activation affecting processes like cell morphology and motility. Furthermore its role extends to insulin signaling where it interacts with components critical in glucose homeostasis.
Pathways
PREX2 engages in the phosphoinositide 3-kinase (PI3K) pathway a vital signaling cascade regulating growth metabolism and apoptosis. PI3K generates phosphatidylinositol (345)-trisphosphate (PIP3) which in turn facilitates PREX2 activation. Through the PI3K pathway PREX2 works alongside proteins like PTEN an important phosphatase that negatively regulates the PI3K pathway balancing cell survival and growth signals. PREX2 also interacts with the mTOR pathway impacting cell proliferation and energy balance.
PREX2 has been linked to cancer and autism spectrum disorders. Its involvement in cancer is noted through its regulation of Rac1 which affects tumor cell motility and invasiveness.
