HINT3 also known as Histidine Triad Nucleotide Binding Protein 3 is a protein encoded by the human HINT3 gene. This protein has a molecular mass of approximately 16 kDa. Mechanically HINT3 functions by binding nucleotides through the histidine triad motif which typically involves AMP and other similar molecules. The expression of HINT3 occurs widely with significant levels found in human brain tissues liver and various other organs which suggests it plays roles in multiple cellular processes.
Biological function summary
The HINT3 protein operates within cells to regulate nucleotide metabolism with implications in various enzymatic activities. It often forms part of larger protein complexes interacting with other nucleotide-binding proteins to execute its functions. Research indicates its involvement in cellular energy homeostasis suggesting a broad impact on maintaining cellular functions by controlling nucleotide turnover and recycling.
Pathways
HINT3 connects to key metabolic pathways like the purine nucleotide biosynthesis and salvage pathways. In these pathways its interactions with related proteins such as IMPDH (Inosine Monophosphate Dehydrogenase) and GMPS (Guanosine Monophosphate Synthetase) highlight its influence in maintaining the balance of nucleotides. These pathways ensure the availability and proper use of nucleotides necessary for DNA and RNA synthesis.
For instance alterations in the expression or functionality of HINT3 might contribute to tumor development as cancer cells often show disrupted nucleotide metabolism. Related proteins such as IMPDH have roles in oncogenesis and their activities can be affected by changes in HINT3 expression or function. HINT3 also shows associations with neurological disorders which are often tied to its expression in brain tissues impacting neuronal nucleotide balance and signaling pathways.
