SAP30 also known as Sin3-associated protein 30 is a component of the Sin3A corepressor complex which modulates gene transcription. This protein has a molecular mass of approximately 30 kDa. SAP30 localizes mainly in the nucleus where it interacts with DNA and other proteins. It is expressed in a wide variety of cell types highlighting its broad regulatory roles in cells.
Biological function summary
SAP30 participates in chromatin remodeling and regulation of gene expression by recruiting histone deacetylases (HDACs) as part of the Sin3A complex. This complex is integral to the repression of transcription during cell cycle regulation and response to DNA damage. By forming a bridge between the Sin3A complex and chromatin SAP30 ensures the recruitment of these enzymes facilitating the removal of acetyl groups from histones which leads to a tighter chromatin structure and repressed gene transcription.
Pathways
SAP30 plays important roles in transcriptional repression pathways and cell cycle regulation. It associates closely with the Sin3A-HDAC complex in these pathways along with proteins such as HDAC1 and HDAC2. The action of SAP30 in these pathways influences cell cycle checkpoints and transcriptional silencing making it an important regulator in maintaining genomic stability and controlling cell proliferation.
SAP30 has associations with cancer and neurodegenerative disorders. Alterations in its regulatory functions can disrupt normal gene expression contributing to tumorigenesis. SAP30 connects with proteins such as p53 and Rb which are often implicated in cancer development. In neurodegenerative disorders the dysregulated expression of genes affected by SAP30 can influence neuronal survival hinting at its potential role in diseases like Alzheimer's where HDAC inhibitors are explored for therapeutic interventions.
