MTGR1 also known as Myeloid Translocation Gene-Related Protein 1 is a transcriptional corepressor involved in gene expression regulation. This protein has an approximate mass of 80 kDa. It is expressed widely in human tissues with higher levels in hematopoietic organs like the bone marrow. MTGR1 interacts predominantly with transcription factors to modulate various biological processes through gene silencing.
Biological function summary
MTGR1 contributes to important cellular processes by acting as a negative regulator of transcription. It forms complexes with transcription factors and histone deacetylases to exert its repressive functions. This protein is a member of the ETO family of corepressors which has several members with distinct but sometimes overlapping roles. Through its interactions MTGR1 influences the transcriptional activity of many genes impacting various cellular functions.
Pathways
MTGR1 participates in the regulation of several transcriptional networks including the Notch signaling pathway. This pathway is essential for cell differentiation proliferation and apoptosis. MTGR1 closely interacts with other coregulators such as MTG8 (ETO) and MTG16 and the AML1-ETO fusion protein providing insights into the pathways’ structural complexity. These interactions highlight its role in fine-tuning the expression of target genes involved in cellular development and maintenance.
MTGR1 plays a role in hematological malignancies such as acute myeloid leukemia (AML). It interacts with proteins like AML1 to form the AML1-ETO fusion protein which is often found in leukemia cases. MTGR1 has also implications in colorectal cancer where its altered expression affects the behavior of tumor cells. These connections underline MTGR1's importance in disease mechanisms and potential as a therapeutic target.
