The FAT atypical cadherin 4 often referred to as FAT4 is a large transmembrane protein with a mass of approximately 560 kDa. It belongs to the cadherin superfamily and contains multiple cadherin repeats EGF-like domains and one laminin G-like domain. FAT4 is expressed widely across tissues with higher expression observed in the kidney liver and cardiovascular system indicating its involvement in diverse physiological processes. This protein plays a role in maintaining cell adhesion and communication.
Biological function summary
FAT4 plays an important role in the regulation of planar cell polarity and tissue organization. It exists within the context of cellular signaling complexes where it functions as a signal transducer that affects cell morphology and motility. Through its interactions with other cadherin family members FAT4 helps coordinate cellular alignment and directional growth. These functions contribute to the proper formation of organ structure and maintain tissue architecture.
Pathways
FAT4 is heavily involved in the Hippo signaling pathway which regulates cell proliferation apoptosis and organ size. It interacts with other proteins like dystrophin and LATS1/2 kinases within this pathway affecting cellular behaviors that are important for development and homeostasis. FAT4 has also been linked to the Wnt signaling pathway where it modulates beta-catenin interactions further linking it to cellular and developmental processes.
FAT4 has connections to Van Maldergem syndrome and certain cancers. Alterations in FAT4 expression or function can lead to defects in tissue formation and cancer progression due to its role in controlling cell growth and polarity. In particular FAT4 mutations have correlated with breast cancer where it interacts with proteins such as YAP and TAZ indicating its potential as a therapeutic target. Understanding FAT4's role in these diseases is key to developing targeted treatments.
