PI-15 also known as Peptidase Inhibitor 15 is a secreted protein characterized by a mass of approximately 12 kDa. This inhibitor is predominantly expressed in the nervous system especially in the human brain and spinal cord. Its primary role involves the regulation of protease activity. The PI-15 protein inhibits trypsin-like serine proteases modulating proteolytic processes that are critical for cellular function.
Biological function summary
PI-15 participates actively in the modulation of protease activity to maintain cellular homeostasis. It does not form parts of larger complexes but exerts its effects individually by inhibiting serine proteases. This inhibition impacts a range of biological processes including cellular differentiation and tissue remodeling. The inhibitory function serves as a regulatory step in proteolytic cascades protecting cells from excessive protease activity that can be damaging.
Pathways
PI-15 integrates into the regulatory landscape of the protease inhibition pathway. Through this pathway it interacts indirectly with proteins such as plasminogen activators contributing to the control of fibrinolysis and matrix degradation. Another pathway involving PI-15 is the extracellular matrix (ECM) remodeling pathway where it helps in managing ECM turnover by inhibiting serine proteases that degrade matrix components maintaining structural integrity.
PI-15 has relevance in neurological conditions and cancer. Its role in the nervous system links it with neurodegenerative diseases possibly affecting the disease process by altering proteostasis. In cancer altered PI-15 expression may influence tumor progression through its effect on ECM remodeling and cellular invasion. The serine proteases targeted by PI-15 including urokinase-type plasminogen activator have also established connections to cancer metastasis emphasizing the importance of PI-15 in cancer biology.
