MC2 also known as adrenocorticotropic hormone receptor (ACTHR) plays a critical role in the adrenal gland's function by binding ACTH. This receptor is a G protein-coupled receptor with a mass of approximately 33 kDa. MC2 expression is localized mainly in the adrenal cortex where it facilitates the regulation of adrenal hormone production essential for physiological stress response.
Biological function summary
The adrenocorticotropic hormone receptor mediates the effects of ACTH by stimulating adrenal steroidogenesis. It serves as part of the MC2 receptor complex which includes the MC2 receptor accessory protein (MRAP). This complex is key in MC2's activation and proper targeting to the cell surface important for cortisol production in response to stress stimuli.
Pathways
MC2 is significantly involved in the hypothalamic-pituitary-adrenal (HPA) axis. This pathway regulates stress-induced hormone release where MC2 interacts with proteins such as MRAP to mediate cortisol secretion. The action of MC2 influences the downstream signaling pathways including the cAMP/PKA pathway which plays a pivotal role in hormone synthesis and secretion regulation.
Alterations in MC2 function are linked to conditions such as familial glucocorticoid deficiency type 1 (FGD1) and Cushing's syndrome. Familial glucocorticoid deficiency arises due to MC2 mutations affecting adrenal response leading to insufficient cortisol levels. The receptor's function interplays with MRAP essential for disease manifestation and a potential therapeutic target. Meanwhile Cushing's syndrome characterized by excessive cortisol may involve MC2 dysregulation in adrenal hyperplasia-related mechanisms.
