RILP known as Rab-interacting lysosomal protein is a critical component in the regulation of membrane trafficking. The protein weighs approximately 45 kDa. RILP assists in tethering and fusion processes essential for vesicle transport acting as an effector for the small GTPase Rab7. It is mainly expressed in the lysosomal compartments of cells. By binding to Rab7 RILP influences endocytic trafficking by coordinating microtubule-based movement of endosomes and lysosomes.
Biological function summary
RILP collaborates within multiprotein complexes influencing lysosomal positioning and maturation. It mediates recruitment of dynein-dynactin motor complexes allowing effective transport of lysosomes along microtubules. Furthermore RILP's activity impacts the degradation pathway assisting in the clearance of cellular debris and turnover of cellular components. It interacts with various cellular structures to maintain the balance between movement and stability of lysosomes which is essential for cellular homeostasis.
Pathways
Researchers place RILP within significant cellular processes like lysosome positioning and autophagic pathways. Its role is important in the endocytic pathway where it acts with proteins like Rab7 and the retromer complex. RILP governs endosome-to-lysosome trafficking impacting cargo sorting and turnover. It shares functional interactions with proteins involved in autophagy influencing the delivery of cellular components for degradation within lysosomes.
RILP notably connects to neurodegenerative diseases and cancers. Altered RILP function can disrupt lysosome positioning and trafficking leading to disorders like Niemann-Pick disease type C. In cancer RILP's interaction with key signaling proteins contributes to tumor progression and metastasis. The protein's regulatory effects on lysosomal pathways intersect with various cellular processes including those involving proteins such as Rab7 that when dysregulated result in pathological conditions.
