Prealbumin also known as transthyretin functions as a transport protein. It has a molecular mass of approximately 55 kDa. This protein primarily expresses in the liver and the choroid plexus of the brain. Prealbumin transports thyroid hormones and retinol-binding protein bound to retinol. MitoNEET is another protein involved in mitochondrial function playing a role in regulating iron-sulfur clusters. Both PIM1 and PIM2 are serine/threonine kinases belonging to the proto-oncogene family with roles in cell survival and proliferation. The expression of PIM kinases varies in tissues but is present in many cell lines.
Biological function summary
Prealbumin transports thyroxine (T4) and triiodothyronine (T3) and plays an important role in thyroid hormone homeostasis. MitoNEET an iron-sulfur cluster protein helps manage oxidative stress in cells. It interacts with various mitochondrial proteins to maintain cellular metabolism. PIM1 and PIM2 kinases act as regulators of cell cycle progression and apoptosis forming a part of oncogenic signaling complexes. These kinases modulate cytokine signaling and immune responses through phosphorylation.
Pathways
Prealbumin is a component of the thyroid hormone transport and metabolism pathway associated with proteins such as albumin and thyroxine-binding globulin. MitoNEET is involved in the cellular response to oxidative stress interacting with proteins like CIAPIN1. PIM kinases act within the JAK/STAT signaling pathway essential for immune function and cell division. PIM1 and PIM2 also closely relate to the PI3K/AKT pathway regulating apoptosis and proliferation through interactions with proteins like BAD and MYC.
Prealbumin relates to amyloidosis and familial transthyretin amyloidosis where misfolded proteins cause organ damage. PIM kinases link to cancers particularly lymphomas and leukemias due to their role in cell survival. PIM1 and PIM2 are associated with tumorigenesis through overexpression leading to increased cell survival and proliferation. MitoNEET connections with metabolic disorders like diabetes arise from its role in mitochondrial metabolic regulation. Its dysfunction contributes to insulin resistance and stress-induced cell damage.
