The target EGFR also known as Epidermal Growth Factor Receptor is a transmembrane protein that plays a critical role in cell signaling pathways involved in cell growth and differentiation. It belongs to a family of membrane-bound receptor tyrosine kinases. EGFR has a molecular mass of approximately 170 kDa and is expressed in various epithelial tissues including the lung liver and pancreas. DNA Polymerase Kappa also referred to as POLK is a specialized DNA polymerase involved in DNA repair with a molecular mass of approximately 98 kDa and it is frequently expressed in the nucleus of multiple cell types.
Biological function summary
EGFR functions by binding to ligands like epidermal growth factor (EGF) leading to its autophosphorylation and the activation of downstream signaling. It participates in several cellular processes such as proliferation migration and survival. EGFR often forms a complex with other HER family receptors influencing the dynamic regulation of signaling processes. In contrast POLK participates in error-prone DNA repair mechanisms allowing the bypass of DNA lesions during replication and it does not typically form such complexes.
Pathways
EGFR is an important player in the MAPK and PI3K/AKT pathways which are important for transmitting extracellular signals that affect cellular responses. It interacts closely with proteins like Ras MEK and Akt within these pathways amplifying diverse cellular responses. On the other hand POLK integrates into the DNA damage tolerance pathway involving proteins such as PCNA and RAD18 which facilitate its role in translesion synthesis during DNA replication.
Aberrant EGFR signaling links to various cancers including non-small cell lung cancer and colorectal cancer. Mutations and overexpression of EGFR can activate oncogenic pathways often in conjunction with proteins like KRAS and BRAF promoting tumorigenesis. POLK's role in inaccurate DNA repair associates with genomic instability which can lead to conditions like cancer. Its interactions with DNA repair proteins including RAD18 can influence the propensity for mutations that drive cancer development.
