TDP1 also known as Tyrosyl-DNA phosphodiesterase 1 plays an important role in DNA repair. This enzyme has a molecular mass of approximately 68 kDa. TDP1 is expressed widely in human tissues predominantly in those with high cellular turnover like bone marrow and the gastrointestinal tract. It mechanistically splits the 3'-phosphotyrosyl bond between DNA and topoisomerase I resolving DNA-Protein cross-links that can otherwise interrupt DNA replication and transcription processes.
Biological function summary
This enzyme is involved in the repair of single-strand breaks by excising the protein residues linked to DNA particularly important for maintaining genome stability. TDP1 forms part of the DNA damage response machinery. In addition to its standalone function it interacts with various other proteins in the DNA repair complex. TDP1 works alongside other repair proteins such as PARP1 to address DNA lesions and maintain cellular integrity.
Pathways
TDP1 significantly contributes to the base excision repair pathway. It aids in counteracting the effects of topoisomerase I inhibitors which are often used in cancer therapy. TDP1 cooperates with SMARCA1 a chromatin remodeler to ensure efficient DNA repair and cellular recovery post-damage. Within this context TDP1's activity is important to both preserving cellular health and conferring resistance to DNA-damaging agents.
TDP1 has been linked to neurodegenerative diseases and certain forms of cancer. Mutations in TDP1 can lead to neurological disorders such as Spinocerebellar ataxia with axonal neuropathy (SCAN1) due to ineffective DNA repair. Additionally cancer cells can exploit increased TDP1 activity to resist chemotherapy highlighting its potential as a therapeutic target. Proteins like PARP1 and topoisomerase I are integral to these disease contexts operating in close association with TDP1 in response to DNA breakage and cellular stress.
