MED13L also known as Mediator Complex Subunit 13-Like is a protein with a molecular mass of approximately 263 kDa. This protein plays a role mechanically as a component of the Mediator complex which is a multiprotein complex that acts as a transcriptional coactivator in eukaryotic cells. MED13L is expressed in various tissues but its expression levels are notably higher in the brain. This indicates its importance in processes related to neural development and function.
Biological function summary
The MED13L protein is integral to the regulation of gene transcription. It functions within the larger context of the Mediator complex which serves as a bridge linking gene-specific transcription factors to the RNA polymerase II enzyme. By modulating the interaction between these transcription factors and polymerase MED13L helps control the expression of target genes. This regulation is important for the proper development and maintenance of many cellular processes particularly in nervous tissues.
Pathways
MED13L is involved in significant biological processes such as cell differentiation and growth. It participates in the Wnt signaling pathway which plays a critical role in embryonic development and cancer pathogenesis. Within this pathway MED13L interacts with other proteins like β-catenin to promote transcriptional activation necessary for cell growth and differentiation. Additionally its role links it to the regulation of the Hedgehog signaling pathway another essential pathway for developmental processes.
Mutations or dysregulation of MED13L have been associated with neurodevelopmental disorders including MED13L haploinsufficiency syndrome which is characterized by intellectual disability and cardiac anomalies. The protein's dysfunction is also linked to certain types of congenital heart defects. In some cases abnormalities in MED13L may interact with components of the Mediator complex affecting the regulation pathways and subsequently contributing to these disorders.
