Dynein intermediate chain 1 also known as DNAI1 is a component of the cytoplasmic dynein complex. Mechanically this protein serves as an adaptor or linker within the larger dynein motor protein complex essential for intracellular transport. Its molecular mass is approximately 74 kDa. DNAI1 is expressed in a variety of tissues with higher expression observed in tissues involved in ciliary function. This protein is important for mediating cargo transport along microtubules contributing to cellular functions such as mitosis endocytosis and organelle positioning.
Biological function summary
Dynein intermediate chain 1 participates in key cellular processes linked to the movement of cilia and flagella. DNAI1 is an integral part of the axonemal dynein complex found in motile cilia. This complex enables the conversion of chemical energy from ATP into mechanical work driving the motion of these hair-like structures. Efficient cilia function relies on the coordinated action of axonemal dyneins enabling the clearance of mucus in respiratory pathways and movement of sperm in reproductive tracts.
Pathways
DNAI1 plays a role in the intraflagellar transport pathway important for building and maintaining cilia. Additionally DNAI1 is involved in the respiratory electron transport chain through its contribution to ciliary movement. The protein interacts closely with dynein heavy chain and dynein light chain proteins within these pathways ensuring the effective transport and movement of cellular components.
Defects in dynein intermediate chain 1 are linked to primary ciliary dyskinesia a genetic disorder affecting ciliary function. This disorder results in chronic respiratory tract infections and reduced fertility due to impaired ciliary motion. DNAI1 mutations can also connect to other ciliary dysfunctions impacting proteins like DNAH5 and DNAH11 which work alongside DNAI1 in the motility of cilia and flagella.
