SENP3 also known as SUMO-specific protease 3 is a protein with a molecular mass of about 66 kDa. It functions primarily as a protease that reverses the SUMOylation of target proteins by cleaving SUMO residues which modulate protein functions and interactions. SENP3 localizes mostly in the nucleoplasm but can also be found in the cytoplasm. The expression of SENP3 varies across different tissues displaying higher levels in the liver heart and skeletal muscle indicating its role in these tissues’ functions.
Biological function summary
SENP3 modulates SUMOylated proteins' activity influencing cellular processes such as transcription signal transduction and stress response. It is part of a larger complex with nucleophosmin a nucleolar protein that assists SENP3 in stability and substrate localization. SENP3's regulation of SUMOylation is critical for maintaining cellular homeostasis and influencing the outcome of signaling pathways related to various stimuli.
Pathways
The activity of SENP3 affects the SUMOylation cycle an important cellular mechanism for post-translational modification. SENP3 plays a role in the DNA damage response pathway where it associates with proteins like NEMO modifying their activity to facilitate repair processes. Additionally SENP3 participates in the oxidative stress response pathway where its action on transcription factors helps cells adapt to stress conditions effectively.
SENP3 is linked to cancer and cardiovascular disease. In certain cancers including hepatocellular carcinoma SENP3 levels are often upregulated leading to altered transcriptional activity and promoting tumor growth. SENP3 also interacts with proteins such as MDM2 influencing p53 degradation and impacting cell cycle regulation. In cardiovascular disease SENP3 contributes to cardiac hypertrophy through its effects on hypertrophic signaling pathways making it a potential target for therapeutic intervention.
