Achaete-scute homolog 3 also known as ASCL3 is a transcription factor with a mass of approximately 24 kDa. This protein belongs to the basic helix-loop-helix (bHLH) family and plays a role in the regulation of neuronal differentiation. ASCL3 shows expression predominantly in neurogenic regions including areas within the brain and peripheral nervous system. It functions by binding to E-box DNA sequences which promotes the expression of genes involved in neurogenesis.
Biological function summary
Achaete-scute homolog 3 drives the differentiation of neuronal precursor cells into mature neurons. It is part of the proneural gene network where it interacts with other bHLH transcription factors to regulate the transcriptional cascade necessary for neuronal development. By forming heterodimeric complexes with E-proteins ASCL3 enhances transcriptional activity thereby promoting the expression of genes critical for neuronal identity and function.
Pathways
Achaete-scute homolog 3 plays a significant role in the Notch signaling pathway which is important for maintaining the balance between neuronal progenitor proliferation and differentiation. It interacts with proteins such as HES1 and HES5 which are also involved in the Notch pathway. By influencing the expression of these pathway components ASCL3 ensures the proper development of neural structures and maintains the equilibrium between different cell fates in neurogenic tissues.
Achaete-scute homolog 3 has been associated with neurodevelopmental disorders. Alterations in its expression or function may contribute to conditions such as schizophrenia and autism spectrum disorders. ASCL3 is connected to other proteins in these conditions such as the DISC1 protein in schizophrenia. Understanding how ASCL3 interacts with these proteins can offer insights into the molecular mechanisms underlying these neurological conditions and potentially guide therapeutic strategies.
