The human multidrug resistance-associated protein 6 (MRP6) also known as ATP-binding cassette sub-family C member 6 (ABCC6) or ARA serves as an important transporter protein. MRP6 weighs approximately 165 kDa and spans the cell membrane to transport various molecules out of the cell. This transport protein expresses highly in the liver and kidney tissues. By exporting certain substrates MRP6 plays an important role in managing cellular environments and protecting tissues from harmful substances.
Biological function summary
MRP6 contributes to the cellular management of organic anions and xenobiotics. It operates as part of the ATP-binding cassette transporters family using energy derived from ATP hydrolysis to drive the transport process. In this role MRP6 can influence the body's detoxification system and help in the excretion of unwanted and potentially toxic compounds. This activity is critical for maintaining homeostasis and protecting cells from damage due to various compounds.
Pathways
Research shows MRP6 operates in the broader context of transporter-mediated drug resistance pathways. Interestingly MRP6 interacts with other proteins such as MRP1 and MRP2 which share similar functions within these pathways. The activity of MRP6 in the extrusion of xenobiotics contributes to a multi-faceted defense mechanism often examined under the scope of chemotherapeutic resistance and transport of physiological metabolites.
Studies highlight MRP6's association with pseudoxanthoma elasticum (PXE) a rare genetic disease characterized by mineralization of elastic fibers in tissues. Mutations in the encoding gene for MRP6 disrupt its functional capacity and lead to disease pathology. Additionally researchers explore the relationship between MRP6 activity and calcification disorders due to its roles in substrate transport and mineral metabolism. MRP6's connection with proteins like ABCC1 emphasizes potential interactions with wider metabolic pathways contributing to disease manifestations.
