Protocadherin 21 also known as PCDH21 or retinal cadherin is a protein with a molecular mass of approximately 101 kDa. This protein is a member of the cadherin superfamily which is known for its role in cell-cell adhesion. PCDH21 is widely expressed in the retina notably in the photoreceptor cells which are critical for visual function. Its presence in these cells points to its significance in maintaining retinal structural integrity and potentially influencing cell signaling pathways necessary for visual processing.
Biological function summary
Protocadherin 21 contributes to the organization and connection of photoreceptors and is an integral component of the photoreceptor sensory cilium complex. This protein plays a role in the development and maintenance of the outer segment discs in photoreceptors which are essential for capturing light and initiating phototransduction. The organization and stability provided by PCDH21 assist in the photoreceptors' effective response to light stimulation emphasizing its importance in normal visual function.
Pathways
Photoreceptor cells rely heavily on the phototransduction cascade to convert light signals into neural signals a process in which Protocadherin 21 plays a supportive role. PCDH21 interacts with other retinal proteins such as rhodopsin which is important for the initiation of this cascade. Additionally links to the Wnt signaling pathway suggest a role in cellular communication and signal transduction highlighting PCDH21's broader interaction within cellular pathways important for proper retinal function.
Protocadherin 21 mutations have connections to inherited retinal diseases such as retinitis pigmentosa and cone-rod dystrophy. These conditions lead to progressive vision loss due to photoreceptor degeneration. Studies show that PCDH21 interacts with proteins like rhodopsin and mutations in either gene may disrupt normal photoreceptor function contributing to retinal degenerative processes. Understanding PCDH21's precise role in these diseases provides promises for targeted therapeutic strategies and better management of vision-related disorders.
