TH1L also known as Mediator Complex Subunit 22 (MED22) is a mechanically important component of the mediator complex a multi-protein structure that acts as a bridge between gene-specific transcription factors and the RNA polymerase II enzyme facilitating the transcription of messenger RNA. TH1L has a molecular weight of approximately 36 kDa. It is predominantly expressed in the nucleus of various cell types often in actively dividing or metabolically active tissues.

The role of TH1L extends beyond just a mechanical scaffold. As part of the mediator complex TH1L plays an important role in regulating gene expression by interacting with transcriptional activators and repressors.
