The Polyribonucleotide 5'-hydroxyl-kinase 3'-phosphatase often known as PNK or PNKP protein is a dual-function enzyme necessary in DNA repair processes. PNKP with a mass of about 57 kDa performs both kinase and phosphatase activities. It removes 3'-phosphates and phosphorylates 5'-hydroxyl groups in polynucleotides preparing them for subsequent repair. This enzyme is expressed in various tissues with significant levels in the brain indicating its widespread importance in cellular processes.

PNKP protein is integral to maintaining genome stability by facilitating the repair of single-strand breaks and double-strand breaks in DNA.
