Abcam's 8 isoprostane competitive
in vitro
ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the determination of 8 isoprostane levels in biological samples. The isoprostanes are a family of eicosanoids of non-enzymatic origin produced by the random oxidation of tissues phospholipids by oxygen radicals. A recent NIH-sponsored study on Biomarkers of Oxidative Stress has indicated that 8 isoprostane is the best index of oxidative injury in a well-accepted oxidant stress rat model. In addition, plasma 8 isoprostane levels were found to be elevated in elderly subjects with severe hypertension and in urine from subjects with high fat diet-induced liver steatosis.
This kit can be used for the determination of 8 isoprostane in diluted urine, serum, plasma, cells and tissues following proper isolation and purification of the eicosanoid from the isoprostane-containing sample.

8-Isoprostane also known as F2-isoprostane is a biomarker used to measure oxidative stress in biological systems. It is a product of lipid peroxidation specifically formed from the non-enzymatic oxidation of arachidonic acid a fatty acid found in the phospholipids of cell membranes. The molecular mass of 8-isoprostane is approximately 352 grams per mole. Researchers commonly find 8-isoprostane in urine and plasma and it is also detected in tissues where oxidant injury is present.
Biological function summary
8-isoprostane is associated with damage signaling due to oxidative stress. It does not form part of larger protein complexes but exists as a standalone molecule derived from lipid peroxidation processes. 8-isoprostane affects the body's responses to oxidative damage and inflammation. It serves as an accurate marker for oxidative stress levels in various conditions due to its relative stability compared to other oxidative stress markers.
Pathways
8-isoprostane plays a role in pathways involving oxidative stress and inflammation. Although it is not part of enzymatic pathways its presence indicates the activation of cellular pathways responding to oxidative damage. 8-isoprostane relates to proteins like cyclooxygenase which also participates in processes connected to inflammation. These pathways highlight the importance of 8-isoprostane in signaling for cellular responses to oxidative injury.
8-isoprostane serves as a marker for conditions involving oxidative stress such as cardiovascular disease and chronic obstructive pulmonary disease (COPD). Its levels correlate with the extent of oxidative damage in these diseases. The assessment of 8-isoprostane can indirectly link to other proteins like protein kinase C which also participates in pathological processes related to oxidative stress and inflammation. This connection makes it a valuable tool in studying the etiology and progression of diseases where oxidative stress plays a significant role.
