Derlin-1 (also known as DERL1 or Derlin-1) is a protein with an approximate mass of 27 kDa. It plays an important role in the endoplasmic reticulum-associated degradation (ERAD) pathway. This protein is involved in the retro-translocation process where misfolded proteins are moved from the endoplasmic reticulum to the cytosol for degradation by the proteasome. DERL1 typically expresses in various tissues with higher levels in secretory cells reflecting its role in protein quality control.
Biological function summary
Derlin-1 helps maintain cellular protein homeostasis and acts as a channel or gatekeeper for misfolded or unfolded proteins. It associates with other ERAD components to form a larger degradation complex. Derlin-1 interacts with proteins such as VCP/p97 and the ubiquitin ligase HRD1 which collaborate to tag defective proteins with ubiquitin preparing them for degradation.
Pathways
The ERAD pathway which Derlin-1 is part of is important for the removal of misfolded proteins protecting cells from stress caused by protein accumulation. Derlin-1's interaction with VCP/p97 and other proteins within the ERAD pathway highlights its function in proteostasis. This process is essential to the Unfolded Protein Response (UPR) another pathway that senses and reacts to misfolded proteins in the endoplasmic reticulum.
Derlin-1's function links to conditions such as cystic fibrosis and certain neurodegenerative diseases. Its role in protein misfolding and degradation relates to these diseases by causing an accumulation of defective proteins. Within the context of cystic fibrosis Derlin-1's role becomes critical in managing faulty CFTR protein degradation. Its interactions with proteins like VCP/p97 can modulate the pathogenesis of neurodegenerative diseases where misfolded protein aggregates play a significant role.
