IL-36 beta also known as IL-1F8 is a member of the interleukin 1 family. It has a mass of approximately 18 kDa. This protein mostly expresses in epithelial tissues including skin and lungs. It acts as a pro-inflammatory cytokine by binding to the IL-36 receptor leading to the activation and recruitment of immune cells to inflammatory sites. This process plays a significant role in host defense mechanisms and wound healing.
Biological function summary
IL-36 beta plays a role in modulating the immune response. It is not part of a complex but acts by enhancing the expression of chemokines and other cytokines. IL-36 beta interacts with inflammatory cells such as T cells and dendritic cells promoting increased inflammation. This mechanistic action makes it an important player in sustaining the inflammatory response and initiating a feedback loop that amplifies its effects.
Pathways
IL-36 beta participates in the NF-kB and MAPK signaling pathways. These pathways are critical to cell survival proliferation and cytokine production. Through these pathways IL-36 beta has interactions with other proteins like IL-1 alpha and IL-1 beta all part of the inflammatory response. This protein network links IL-36 beta to wider immune system activities and cellular responses to stress.
IL-36 beta is strongly associated with psoriasis and rheumatoid arthritis. In these conditions the overexpression of IL-36 beta can result in chronic inflammation. In psoriasis IL-36 beta works closely with proteins like IL-17A and TNF-alpha which also contribute to the pathogenesis of the disease. Similarly in rheumatoid arthritis increased IL-36 beta levels correlate with inflammation and joint damage establishing its role in exacerbating disease symptoms.
