How the assay works

This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. As an example, the membrane form of TNF-alpha appears to mediate anti-tumorigenic therapeutic responses whereas the soluble ligand is linked to inflammation and proliferation.

Tumor necrosis factor alpha (TNF alpha) also known as TNF-a is a cytokine with a molecular weight of approximately 17 kDa. This protein is expressed mainly by macrophages though other cells like T-cells and natural killer cells also produce it. TNF alpha helps in regulating immune responses and inflammatory reactions. Its expression is important in triggering cell apoptosis and modulating immune cell activities. TNF alpha exists in membrane-bound and soluble forms both having biologic activity.
Biological function summary
TNF alpha acts as an important regulator of inflammatory and immune responses. It is not part of a complex but interacts with TNF receptors to activate various signaling cascades. TNF alpha promotes leukocyte recruitment to sites of infection or injury by enhancing adhesion molecule expression on endothelial cells. Additionally it induces the production of other pro-inflammatory cytokines like IL-6 and IL-1 amplifying the immune response.
