Hepatocyte nuclear factor 6 (HNF6) also known as Onecut1 is a transcription factor that functions to regulate gene expression. It has a molecular mass of approximately 55 kDa. HNF6 is expressed in the liver pancreas and several other tissues influencing the development and function of these organs. It plays a significant role in regulating genes involved in various cellular processes by binding to DNA sequences and controlling transcription.
Biological function summary
This transcription factor controls the development and maturation of pancreatic islets and hepatocytes. It does not usually function as part of a larger protein complex but it directly influences genes that are vital for these processes. HNF6 ensures proper liver cell differentiation and has an important role in managing glucose metabolism and bile acid homeostasis in tissues where it is expressed.
Pathways
Hepatocyte nuclear factor 6 is integral to the glucose metabolism and bile acid biosynthesis pathways. It interacts with other transcription factors such as HNF4α and FOXA2 forming a regulatory network. HNF6 affects gene expression involved in maintaining metabolic balance and liver detoxification processes illustrating its critical influence on metabolic function within these pathways.
HNF6 links to diabetes and liver diseases. Its modulation of pancreatic development and function implicates it in both type 1 and type 2 diabetes often involving its interaction with other transcription factors like PDX1 and MafA. Similarly changes in HNF6 expression affect liver diseases such as cholestasis where it impacts bile acid regulation and liver cell function.
