Swi6 also known as Char9 or Split Ends is a protein recognized for its role in cell cycle regulation. This protein has a mass of approximately 89 kDa and primarily expresses in the nucleus of both yeast and certain eukaryotic cells. Swi6 functions as a regulatory subunit and plays an integral role in transcription regulation. Swi6 also contributes to modulating the timing and coordination of critical cell cycle events.
Biological function summary
Swi6 binds with transcription factors and contributes to the formation of cell cycle-dependent kinase complexes. It assists in the regulation of transcription when it acts as a part of the SBF and MBF complexes. These complexes are responsible for the transcriptional activation of genes necessary for G1/S transition during the cell cycle. The ability of Swi6 to selectively bind to specific DNA sequences makes it fundamental in controlling gene expression patterns.
Pathways
Swi6 operates within the cell cycle control pathways. It interacts with the retinoblastoma (Rb) protein to influence the G1 checkpoint control pathway. In yeast Swi6 is also part of the START checkpoint pathway ensuring proper DNA synthesis initiation. Its association with proteins like Cln3 and Cdc28 is central to its function in these pathways linking external growth cues to internal cell cycle progression mechanisms.
Swi6 impacts cancer progression and development. Its involvement with cell cycle regulation connects it to various cancer types. Aberrant expression or mutations in Swi6 can lead to dysregulation of cell division often associated with the development of malignant cells. The protein's interaction with the Rb protein is also significant in cancer biology as disruptions in this pathway are frequently observed in tumorigenesis notably in retinoblastoma and other cancers.
