E-Syt1 also known as Extended Synaptotagmin 1 is a protein encoding a molecule of approximately 129 kDa. This protein plays an essential role in the tethering of the endoplasmic reticulum (ER) to the plasma membrane facilitating lipid transfer between these organelles. It is mostly expressed in the brain and other tissues. E-Syt1 possesses N-terminal transmembrane segments that anchor it within the ER membrane and its C-terminal part reaches out to bind PI(45)P2 at the plasma membrane upon increased cytosolic calcium levels.
Biological function summary
The Extended Synaptotagmin family which includes E-Syt1 forms a complex known as the E-Syt1/SMP complex which aids in the transfer of lipids for membrane repair and cellular homeostasis. The protein ensures cell membrane integrity by shuttling lipids in response to cellular signals. This mechanism supports cell survival and function especially under stress conditions where membrane damage may occur.
Pathways
E-Syt1 is integral to phosphoinositide and calcium signaling processes. It forms important interactions within the phosphoinositide metabolism pathway facilitating membrane dynamics. The protein also intersects with pathways involving calcium signaling due to its calcium-binding capacity working alongside proteins like Synaptotagmin to fine-tune cellular responses to fluctuations in calcium levels.
Extended Synaptotagmin 1 has implications in conditions like neurodegenerative diseases and certain cancers. Alterations in E-Syt1 activity or expression levels can contribute to conditions such as Alzheimer’s disease where disrupted lipid transport affects neuronal health. E-Syt1 in tandem with proteins like tau in Alzheimer’s highlights its role in maintaining cellular functions that prevent disease progression. Its involvement in lipid transfer can also relate to cancer pathologies where cell membrane dynamics are important.
