The ING3 protein also known as inhibitor of growth family member 3 is a nuclear protein with a molecular mass of approximately 47 kDa. ING3 belongs to the ING gene family and acts as a tumor suppressor by modulating chromatin structure and regulating gene expression. It is expressed in various tissues including the liver kidney and heart and plays a critical role in cellular stress responses. ING3 interacts with several components of the histone acetyltransferase (HAT) complex regulating processes like cell growth apoptosis and DNA repair.
Biological function summary
ING3 influences gene transcription and chromatin remodeling as a part of the NuA4 histone acetyltransferase complex. This complex is involved in the acetylation of histone H4 and H2A facilitating an open chromatin structure that permits transcriptional activation. ING3 is important in the regulation of cell cycle progression and apoptosis. It modulates p53 and other key cellular pathways underpinning its role in maintaining normal cellular homeostasis and preventing cancerous transformations.
Pathways
ING3's function supports significant biological processes like DNA repair and apoptosis. It is part of the p53 signaling pathway which plays a pivotal role in the cellular response to DNA damage. ING3 acts alongside proteins such as p21 and MDM2 in the p53 pathway. Furthermore ING3 interfaces with the MAPK/ERK signaling pathway contributing to its regulation of cell division and survival. Through these pathways ING3 helps orchestrate a balanced response to cellular stress and damage.
Researchers associate ING3 with cancers particularly breast and liver cancers. Studies indicate that downregulation or mutation of ING3 leads to disrupted cell cycle control and increased tumorigenesis. Furthermore ING3's interaction with proteins such as p53 emphasizes its critical role in tumor suppression. By modulating pathways linked to proliferation and apoptosis ING3 provides potential targets for therapeutic intervention in cancer treatments highlighting its relevance in disease management and research.
