CTPS2 or CTP synthase 2 plays a role in pyrimidine biosynthesis by converting UTP to CTP an essential step in nucleotide metabolism. This enzyme has a mass of approximately 64 kDa. CTPS2 is expressed in various tissues with notable expression in immune-related cells and the liver. Its activity contributes to maintaining a balance between the pyrimidine nucleotides necessary for DNA and RNA synthesis.
Biological function summary
CTPS2 is involved in regulating nucleotide pools which is critical for cell proliferation and differentiation. This protein does not typically form part of a larger complex but acts as a homotetramer. CTPS2's function ensures that cells have enough cytidine triphosphate (CTP) for various cellular processes including lipid synthesis and glycosylation reactions vital for maintaining cell membrane integrity and glycoprotein functions.
Pathways
CTPS2 plays a significant role in the pyrimidine metabolic pathway. It is directly involved in the de novo synthesis of CTP. This pathway links to the salvage pathway where it interacts indirectly with proteins involved in nucleotide salvage like uridine-cytidine kinase (UCK) which phosphorylates nucleosides to monophosphate nucleotides. Through these pathways CTPS2 influences DNA replication and cell growth.
CTPS2 has connections to cancer and immunodeficiency disorders. Overexpression of CTPS2 is seen in certain cancers where it may contribute to unchecked cell division. On the other hand impaired CTPS2 function can result in reduced immune response affecting T cell proliferation. The protein is also linked to ribonucleotide reductase M2 (RRM2) through pathways impacting the balance of deoxyribonucleotides in rapidly dividing cells.
