The IRE1β also known as ERN1B is a transmembrane endoribonuclease with a mass of approximately 110 kDa. It belongs to the IRE1 family and acts as an important sensor of the unfolded protein response (UPR) pathway. IRE1β is expressed mainly in epithelial cells of the intestine with some presence in lungs and liver tissues. The protein comprises a luminal domain that detects misfolded proteins along with a kinase and RNase domain that initiates downstream signaling pathways.
Biological function summary
IRE1β plays a significant role in cellular stress responses by regulating mRNA splicing and degradation especially during endoplasmic reticulum (ER) stress events. It operates as a part of a complex signal transduction mechanism that maintains cellular homeostasis under stress. In particular upon activation IRE1β engages in unconventional splicing of the transcription factor XBP1 mRNA which in turn upregulates genes involved in protein folding and ER-associated degradation.
